摘要
目的:探讨细粒棘球蚴(Echinococcus granulosus,Eg)囊液(HCF)对大鼠肝星状细胞(HSC)T6细胞系增殖的影响。方法:HCF体外刺激T6细胞,通过MTT法检测细胞增殖,流式细胞仪检测细胞周期,Western Blot检测细胞外调节激酶(ERK1/2)磷酸化、细胞增殖核抗原(PCNA)及细胞周期蛋白(cyclin)D1、E、A和B1表达水平。结果:6 h和12 h,15%、30%和60%HCF组与对照组相比增殖显著(P<0.05);15%HCF组细胞增殖指数分别为57.9%和71%;Western Blot检测显示15%HCF组pERK、PCNA和cyclin E在0.5 h时的表达量较为对照组差异显著(P<0.05);cyclin D1在6 h时表达量较对照组差异显著(P<0.05)。结论:HCF可能是通过激活ERK后调控细胞周期相关蛋白的表达促进了HSC的增殖。
Objective: To discuss the effect of Echinococcus granulous cyst fluid on mitogen - activated protein kinases (MAPK) and the cell cycle. Method:T6 cells cultured with different concentrations of hydatid cysttluid (HCF) were tested by the MTT method to determine effects on proliferation. The cell cycle was assessed by flow eytometry. Western Blot indicated the expression of p-ERK, PCNA, cyclinD1, cyclinE,eyclinA and cyclinB1. Result:6 h and 12 h of HCF at 15% ,30% and 60% HCF concentrations in the cell media signification promoted cell proliferation and compared with controls. The flow cytometry analysis showed the HCF accelerated cell cycle progression; Cell exposed to 15% HCF for 0. 5 h showed significantly induced expression of p - ERK,PCNA and cyelin E higher than controls (P 〈 0. 05). Cell exposed to 15% HCF for 6 h showed significantly induced expression of cyclin DI, higher than controls (P〈0. 05). Conclusion:HCF promoted the proliferation of rat hepatic stellate eeUs may be by activation of ERK and regulate the expression of cell cy-cle - related proteins.
