缝隙连接抑制剂辛醇对脑缺血不同时间再灌注大鼠脑梗死体积和神经元损伤的影响 被引量:2

摘要 目的探讨缝隙连接抑制剂辛醇对脑缺血不同时间再灌注大鼠脑梗死体积和神经元损伤的影响。方法 96只大鼠随机分为脑缺血对照组和辛醇干预组,每组又分为缺血30 min亚组和缺血2 h亚组。采用改良的zea-Longa线栓法制作局灶性脑缺血大鼠模型。辛醇干预组于术前30 min按5 mmol/kg的剂量腹腔注射辛醇。脑缺血对照组予以等量二甲基亚砜溶液替代。各组缺血相应时间(30 min或2 h)后再灌注24h。采用氯代三苯基四氮唑染色检测脑梗死体积,苏木精-伊红染色观察脑组织神经元损伤。结果脑缺血对照组缺血30 min亚组的脑梗死体积及神经元损伤明显小于辛醇干预组缺血30 min亚组(均P<0.05);而脑缺血对照组缺血2 h亚组的脑梗死体积及神经元损伤明显大于辛醇干预组缺血2 h亚组(均P<0.05)。结论缝隙连接抑制剂辛醇对局灶性脑缺血再灌注损伤存在双向作用,且可能与脑缺血时间有关。 Objective To investigate the effects of gap junctional blocker octanol on infarction volume and neuronal injury in ischemia reperfusion rats with different ischemia time. Methods Ninty-six rats were randomly divided into ischemia group and octanol group, which were further divided into ischemia 30 min and 2 h subgroups. Focal cerebral ischemic rat model was induced by intraluminal suture technique modified by zea-Louga. Octanol group was administrated intraperitoneally 30 rain before ischemia at the concentration of 5 mmol/kg. Ischemia group received equal volume of dimethyl sulfoxide. Each group was reperfused for 24 h after occluded for 30 min or 2 h. Infarction volume was compared by 2,3,5-Triphenyhetrazolium chloride staining and pathological changes were observed by hematoxylin-eosin staining. Results The infarction volume and neuronal damage in ischemia 30 rain subgroup of ischemia group were significantly less than that in ischemia 30 rain subgroup of octanol group ( all P 〈 0. 05 ). The infarction volume and neuronal damage in ischemia 2 h subgroup of ischemia group were markedly larger than that in ischemia 2 h subgroup of octanol group ( all P 〈 O. 05 ). Conclusion Gap junctional blocker oetanol played a bidirectional role in focal ischemia reperfusion, which may be related with the ischemia time.
