摘要 目的探讨神经胶质瘤患者miRNA-193b基因表达水平的改变及其意义。方法对72例神经胶质瘤患者(神经胶质瘤组)和30例脑外伤手术患者(对照组)的脑组织标本进行miRNA-193b基因表达水平的检测。采用甲基化特异性PCR技术对神经胶质瘤患者miRNA-193b启动子区甲基化水平进行检测。对结果进行比较分析。结果对照组miRNA-193b基因表达水平(0.5528±0.1957)明显高于神经胶质瘤组(0.2450±0.1845)(P<0.01)。低级别亚组神经胶质瘤患者miRNA-193b基因表达水平(0.2909±0.1802)明显高于高级别亚组(0.1991±0.1795)(P<0.05)。低甲基化亚组神经胶质瘤患者miRNA-193b基因表达水平(0.2817±0.1883)明显高于高甲基化亚组(0.1615±0.1476)(P<0.01)。结论神经胶质瘤患者miRNA-193b基因表达水平低,且病理级别越高、甲基化水平越高,miRNA-193b基因表达水平越低。该基因表达水平可作为神经胶质瘤早期诊断或病情监测的指标之一。 Objective To explore the change and its significance of the expression level of miRNA-193b gene in patients with glioma. Methods The expression levels of miRNA-193b gene in brain tissues of 72 patients with glioma (glioma group) and 30 cerebral trauma patients with operated (control group) were detected. The methylation level at promoter region of miRNA-193b gene in patients with glioma was detected by methylation-specific PCR. The results were compared and analyzed. Results The expression level of miRNA-193b gene in control group (0.5528 _+0. 1957 ) was significantly higher than that in glioma group (0. 2450 -+ 0. 1845 ) ( P 〈 0. 01 ). The expression level of miRNA-193b gene in low degree subgroup of glioma patients (0. 2909 _+ 0. 1802) was significantly higher than that in high degree subgroup (0. 1991 -+ 0. 1795) (P 〈 0. 05). The expression level of miRNA-193b gene in low methylation level subgroup of glioma patients (0. 2817 ~: 0. 1883 ) was significantly higher than that in high methylation level subgroup (0. 1615 + 0. 1476) (P 〈 0.01 ). Conclusion The expression level of miRNA-193b gene in patients with glioma is low, and the higher of the pathologic degree and methylation level, the lower of the expression level of miRNA-193b gene, which can be one of the indexes for early diagnosing or illness monitoring.
