摘要
抗体药物属于生物大分子药物,其生物功能的发挥离不开复杂的翻译后修饰。糖基化修饰作为抗体最重要的翻译后修饰,对于抗体的生物活性和体内代谢有着重要的作用。本文综述了已上市治疗性抗体的主要糖基化类型与分析方法,并对不同工程细胞株产生抗体的糖基化修饰进行了比较。鉴于不同的糖基化对于抗体亲和力和体内代谢具有显著影响,以功能研究为基础的糖基化工程已广泛应用于新型抗体的研发。
Therapeutic antibodies, belonging to biological macromolecule, exert their pharmaceutical functions related to completed post - translational modification. As one of the key modification , glycosylation significantly influences the activity and metabolism of antibodies. Here, we summarized the main glycosylation types and analysis methods of therapeutic antibody, and compared the glycosylation between antibodies produced in different cell types. Distinct glycosylation could lead to various affinity and metabolism manners of the therapeutic antibodies, thus the antibody Glyco - engineering based on functional study has been widely applied in the development of novel therapeutic antibodies.
