摘要
目的:通过生物信息学方法分析人巨细胞病毒包膜糖蛋白B(gB)的三级结构并预测其线性B细胞抗原表位.方法:根据人巨细胞病毒gB蛋白的胞外域序列,利用DNAStar软件分析其二级结构和表面特性,联合在线B细胞表位预测程序BcePred,通过其理化特性来综合预测该蛋白可能的线性B细胞抗原表位;利用SWISSMODEL服务器的同源建模方法构建gB蛋白的三级结构模型,对在生理三聚体条件下该蛋白各预测表位进行验证.结果:成功预测出人巨细胞病毒gB蛋白的多个线性B细胞抗原表位区段,其中预测出的5个表位区段不属于任何已发现的抗原区域,为后续验证其优势中和表位,研制安全、高效的表位疫苗奠定了理论基础.
Objective: Using bioinformatics approaches, the B cell linear epitopes of Human Cytomegalovims GlycoproteinB (gB) are predicted by studying its physicochemical properties. Methods: Based on the sequence of the HCMV gB extracellular portion, the probable B cell epitopes of gB are analyzed with its secondary structure and surface properties through DNAstar software as well as the BcePred online program. Meanwhile, the tertiary structure of gB is constructed by homologous modelling with the assistance of SWISSMODEL server, analyzing the surface possibility of epitopes under the condition of physiological trimer. Results: This study successfully predicted several linear B cell epitopes of the extracellular portion of HCMV gB, in which 5 epitopes were reported for the first time. And this work could provide a theoretical foundation for further verification of the gB immunodominant epitopes and developing safe and effective epitope vaccines.
