摘要
目的:探讨雷公藤多甙对核因子的影响并为药物治疗银屑病提供理论依据。方法:密度梯度离心法获得外周血单一核细胞(PBMC),体外培养角质形成细胞(KC)。PBMC与KC混合培养,流式细胞仪检测核转录因子κB(NF-κB)表达,ELISA法测上清液白介素-8(IL-8)、细胞间黏附分子-1(ICAM-1)含量。结果:银屑病组NF-κB表达及ICAM-1、IL-8含量与正常对照组间差异有显著性(P<0.01);雷公藤可下调NF-κB表达及IL-8、ICAM-1含量(P<0.01)。结论:雷公藤可能通过影响NF-κB的表达而发挥对银屑病的治疗作用,抑制NF-κB的药物可能有助于银屑病的治疗。
Objective: To explore the effects ofglucosidorum tripterygii tororum on nuclear factor and provide theoretical evidence for treating psoriasis with drugs. Method: Peripheral blood mononuclear cells (PBMC) were obtained by density gradient centrifugation, and keratinocytes (KC) were cultured in vitro, PBMC and KC were mixed and cultured, the expressions of NF-KB were detected with flow cytometry and the contents of ICAM-1 and IL-8 in the liquid supernatant tested with ELISA method. Result: Psoriasis group showed significant difference compared with the normal control group in the expressions of NF-KB, the contents of ICAM-1 and IL-8(P〈0.01); LeiGongTeng (tripterygium wilfordii) could lower the expressions of NF-κB and the contents of IL-8 and ICAM-1 (P〈0.01). Conclusion: LeiGongTeng could improve the symptoms of psoriasis through influencing the expressions of NF-KB, drugs of inhibiting NF-KB might be helpful to the treatment of psoriasis.
