大鼠脂肪源性干细胞的分离与鉴定 被引量:1

摘要 目的:体外分离培养大鼠脂肪源性干细胞,观察并记录大鼠脂肪源性干细胞的生长特点,探索其成骨分化潜能。方法:取SD大鼠腹股沟脂肪垫,消化分离出脂肪源性干细胞后进行传代培养。用MTT(四甲基偶氮唑蓝)法对原代及1,3,5,7,9,11,13代细胞进行生长测定。流式细胞仪下对第4代脂肪源性干细胞进行表面标记物CD44,CD106,CD105,CD49d的表达测定。取第3代脂肪源性干细胞进行体外诱导分化实验:地塞米松、β-甘油磷酸钠、维生素C作用下成骨方向诱导。结果:细胞表面标记物CD44,CD106,CD105,CD49d阳性率分别为99.59%,0.85%,95.00%,89.56%。MTT绘制生长曲线显示经多次传代后细胞仍能保持较强的增殖能力。成骨诱导组诱导后,茜素红染色下可见钙结节沉积。结论:大鼠脂肪源性干细胞,持续至第13代仍然能够保持旺盛的分裂增殖能力及特征,在特定的条件下可分化为成骨细胞。
