电化学发光免疫传感器检测癌胚抗原

摘要 以鲁米诺还原纳米银作为信号分子标记癌胚抗原(CEA),结合具有良好生物相容性的纳米金,固定癌胚抗原抗体于金电极上,成功建立了用于检测人血清中CEA含量的电化学发光(ECL)免疫分析方法。利用该方法可在浓度为0.1-50ng·mL-1的线性范围内稳定监测人血清中CEA的含量,检测限为0.05ng·mL-1。将该电化学发光传感器应用于健康志愿者的血清中进行CEA的检测,结果令人满意。相对标准偏差小于7.2%,平均回收率为97.2-102.9%,具有良好的稳定性和重现性。 Electrochemiluminescence immunoassay for the determination of carcinoembryonic antigen (CEA) was created with nanosilver as the signal molecule labeled on CEA and with nanogold to fix anti- CEA on gold electrode. This method can be used to determine the content of CEA in human serum within a linear range of 0.1-50ng.mL-1 with a LOD (limit of detection ) of 0.05ng-mL^-1. The electroehemiluminescence immunosensor has been successfully applied in the determination of CEA in the serum of healthy volunteers. The RSD has been lower than 7.2%, and the average recovery rate has been 97.2-102.9%, which shows the sensor's good stability and reproducibility.
