摘要
目的明确大鼠蛛网膜下腔出血(subarachnoid hemorrhage,SAH)后皮层扩散性抑制(cortical spreading depression,CSD)对皮层脑血流及细胞凋亡的影响。方法采用枕大池2次注血法建立大鼠SAH模型,利用皮层给以KCl的方法诱导CSD的发生,激光多普勒血流仪行皮层脑血流测定,TUNEL染色检测细胞凋亡。结果 SAH组大鼠脑皮层血流较空白组和盐水组明显下降,其中5d组最低;皮层细胞凋亡水平明显升高,5d组最明显;CSD发生后SAH组脑血流下降更明显,凋亡更严重,空白组和盐水组变化不明显。结论 SAH后CSD的发生使脑血流进一步下降,皮层细胞的凋亡更加严重,CSD可能是导致迟发性缺血性神经功能障碍的机制之一。
Objective To clarify the effect of cortical spreading depression (CSD) on cortical cerebral blood flow and apoptosis following subarachnoid hemorrhage (SAH) in rats. Methods SAH was induced in male Wistar rats by autologous blood injection into the cisterna magna and CSD was induced in rats with KC1. The cortical blood flow was measured by laser Doppler flowmeter and apoptosis were detected by TUNEL staining. Results In SAH groups the cortical cerebral blood flow was significantly reduced and cortical cell apoptosis was increased compared with those in blank and normal saline groups. After CSD, the cortical cerebral blood flow and apoptosis in SAH groups were more obvious than in blank and normal saline groups. Conclusion CSD can reduce cortical cerebral blood flow and aggravate apoptosis after SAH. It may be one of the mechanisms of delayed ischemic neurologic deficit.
