摘要
[目的]筛选金银花试管苗的最佳生根培养基。[方法]以金银花的腋芽为外植体,以萘乙酸(NAA)、吲哚丁酸(IBA)和多效唑(MET)为考察因素,通过正交试验优选金银花试管苗的生根培养基。[结果]激素对生根率影响的大小依次为NAA>IBA>MET;以生根率为指标的最佳培养基为B5+0.05 mg/L NAA+0.2 mg/L IBA+1.0 mg/L MET。激素对主根数影响的大小依次为IBA>NAA>MET;以主根数为指标的最佳培养基为B5+0.4 mg/L IBA+0.2 mg/L NAA+0.5 mg/L MET。激素对须根数影响的大小依次为NAA>IBA>MET;以须根数为指标的最佳培养基为B5+0.05 mg/L NAA+0.4 mg/L IBA+0.5 mg/L MET。[结论]该研究筛选了金银花试管苗的最佳生根培养基,为金银花试管苗的工厂化育苗提供了基础。
[ Objective ] To screen the optimal rooting medium of Lonicera japonica test-tube plantlet. [ Method ] The axillary bud of Lonicera ja- ponica was used as explant and the effects of different hormones ( NAA, IBA, MET) on rooting were studied through the orthogonal test. [ Resuit] The effects of different hormones on rooting radio was NAA 〉 IBA 〉 MET, the optimal medium of rooting radio was B5 + NAA 0.05 mg/L + IBA 0.2 mg/L + MET 1.0 mg/L. The effects of different hormones on the main root amount was IBA 〉 NAA 〉 MET, the optima medium of the main root amount was B5 + IBA 0.4 mg/L + NAA 0.2 mg/L + MET O. 5 mg/L. The effects of different hormones on the fibrous root amount was NAA 〉 IBA 〉 MET, the optima medium of fibrous root amount was B5 + NAA 0.05 mg./L + IBA 0.4 mg/L + MET 0.5 mg/L. [ Conclusion] The optimal rooting medium for Lonicera japonica test-tube plantlet was obtained, which will provide a basis for the industrial seedling of Lonicera ja- ponica.
