高效液相色谱法测定黏膜溃疡脂中醋酸地塞米松的含量

Determination of Dexamethasone Acetate in Ulcer Grease by HPLC

目的 建立高效液相色谱法测定黏膜溃疡脂中醋酸地塞米松的含量.方法 采用高效液相色谱法,色谱柱为Sunfire C18柱（4.6×250mm,5μm）;流动相为乙腈：水（70：30）;流速为1.0mL﹒min-1;柱温30℃;检测波长为240nm.结果 醋酸地塞米松的质量浓度在5.43～86.88μg·mL-1范围内呈良好的线性关系（r=0.9998,n=5）,平均回收率为98.9%（RSD=1.26%,n=9）.结论 本法操作简单准确,重现性好,为控制黏膜溃疡脂的质量提供了良好的依据.

OBJECTIVE To establish a method-for the determination of dexamethasone acetate in ulcer grease by HPLC. METHODS A new quantitative analysis of dexamethasone acetate in ulcer grease was adopted by HPLC. The column was Sunfire C18 （4.6×250mm,5μm）, with mobile phase of acetonitrile-water（70：30）at the flow rate of 1.0mL·min -1, the column temperature was 30℃. Detection wavelength was 240nm. RESULTS The method had good inear relationship within the range of 5.43 - 86.88 μg·mL-1（ r = 0. 9998） for dexamethasone acetate（r =0. 9998,n =5） ,the average recoveries（n =9） was 98.9% （ RSD = 1.26% ,n =9）. CONCLUSION The method is well-done for the determination of dexamethasone acetate. It can be used for quality control of ulcer grease.
