大鼠脑缺血再灌注后Bcl-2和Bax的表达

摘要 目的探讨大鼠脑缺血再灌注(CIR)后早期凋亡相关基因Bcl-2和Bax在脑皮质神经元中的表达变化。方法将56只大鼠随机分为假手术组(对照组)及缺血再灌注组(观察组),采用免疫组织化学法测定Bcl-2和Bax的表达。结果随着CIR时间的延长,凋亡细胞增多,缺血组Bcl-2、Bax表达增多,与对照组相比,差异有统计学意义(P<0.01),Bcl-2的表达在12h达到最高,Bax的增加一直持续到48h。结论 CIR后Bcl-2表达的增加早于Bax,CIR后神经细胞凋亡可能与Bcl-2/Bax失调有关。 Objective To investigate the changes ofBcl-2, Bax and caspase-3 expression in cerebral cortical neurons after cerebral ischemia-reperfusion (CIR). Methods 56 rats were randomly divided into sham operation group (control group) and ischemia-reperfusion group (observation group); the changes of Bcl-2 and Bax expressions were determined by immunohistochemistry. Results With the extension of CIR time, compared with control group, the apoptotic cells increased, and Bcl-2, Bax expressions increased in ischemic group, the difference was statistically significant (P〈0.01); Bcl-2 expression reached a peak value at 12h post-reperfusion, while Bax increased until 48h post-reperfusion. Conclusion The expression of Bcl-2 in cerebral cortical neurons increased earlier than Bax after CIR, suggesting neuronal apoptosis caused by CIR may be associated with Bcl-2/Bax ratio disorders.
