协同刺激分子CD46诱导T细胞分泌IL-10的研究 被引量:1

摘要 目的探讨补体调节蛋白CD46作为一种新的T细胞共刺激分子参与T细胞活化的作用机制。方法磁珠分离纯化人外周血中CD4+T细胞,在加入或未加入外源性IL-2情况下,采用CD3/CD46或CD3/CD28刺激培养。ELISA方法测定培养上清液中IL-10、IFN-γ及IL-4的浓度,用氚标记胸腺嘧啶核苷(3H-TdR)掺入法测定细胞增殖速率。结果在加入外源性IL-2时,CD3/CD46或CD3/CD28刺激CD4+T细胞分泌IL-10水平较缺乏IL-2时明显增加。在中等浓度CD3刺激下,以CD46为协同刺激分子时,CD4+T细胞分泌IL-10水平较以CD28为协同刺激分子时水平升高。以CD46为协同刺激分子时,CD4+T细胞分泌IFN-γ水平降低,两者均无IL-4分泌。在CD3/CD46刺激下,CD4+T细胞增殖能力较CD3/CD28刺激下显著降低。结论 CD46作为一种新的T细胞共刺激分子能够刺激T细胞活化,并导致IL-10分泌增加。 Objective To study the mechanism of T cell activation mediated by complement regulatory factor CD46 which provides a eo-stimulatory signal. Methods Purified CD4^+ T cells were cultured in the presence or absence of IL-2 under stimu-lation with CD3/CD46 or CD3/CD28. The levels of IL-10,IFN-γ and IL-4 in supernatants were measured with ELISA;prolifera-tion rates were assayed by(3H-TdR)thymidine incorporation. Results In the presence of IL-2,1evels of IL-10 were higher than in absence of IL-2 under stimulation with CD3/CIM6 or CD3/CD28. When stimulated with middle lever of CD3 ,the lever of IL- 10 was higher under CD46 eostimulation than under CD28 costimulation;the lever of IFN-γ, was lower under CD46 costimulation and no IL-4 were tested under neither of situation. The proliferation rates were lower under CD46 costimulation. Conclusion CD46 as a new co-stimulatory signal can activate T cells and induce higher IL-10 secretion than traditional signal.
