射频消融对消融灶边缘肿瘤细胞生长的影响

摘要 目的探讨射频消融(radiofrequency ablation,RFA)在体外细胞实验中对消融灶边缘残余肿瘤细胞生长的影响。方法使用锥虫蓝染色法观察不同温度和时间RFA作用后HCT116细胞的生长情况,以及不同温度RFA作用后消融灶边缘HCT116细胞在不同时间点的生长情况;利用流式细胞仪检测不同温度RFA后24 h消融灶边缘HCT116细胞的凋亡率。结果 RFA后,HCT116细胞从射频中心向外依次形成凝固性坏死区域、水泡样变条带和未发生明显变化的区域。随着作用温度的升高和作用时间的延长,锥虫蓝的蓝染区域扩大,蓝染区域周围出现细胞脱落带。85℃、70℃和55℃RFA后24 h,消融灶周围残余HCT116细胞的凋亡率分别为(18.34±0.94)%、(10.89±0.77)%和(11.90±1.33)%,均明显高于空白对照组(2.45±0.09)%(P<0.01)。85℃组细胞的凋亡率显著高于70℃组和55℃组(P<0.05)。结论体外实验发现,RFA可对肿瘤细胞产生进行性损伤,同时可促进消融灶周围残余肿瘤细胞凋亡。 Objective To investigate the effect of radiofrequency ablation (RFA) on the growth of the residual tumor cells around the focus of RFA in vitro. Methods Trypan blue staining was applied to observe HCTll6 cell death after RFA at various temperatures and times, and the growing status of the residual HCT116 cells around the focus of RFA at different time points after various temperatures RFA. Then flow cytometry was used to detect the apoptosis rates of the residual HCT116 cells around the focus of RFA after variant temperatures of RFA. Result After RFA, HCT116 cells from the focus of RFA to the peripheral area sequentially formed the coagulative necrosis area, bubble-like cells loop, and the area with no remarkable changes of the ceils. As applying temperature rose and time extended, the blue-stained area expanded, and the cells in the loop around the blue-stained area began to fall off. Twen- ty-four hours after 85℃, 70℃ and 55℃ RFA , the apoptosis rates of residual HCT116 ceils around the focus of RFA were respectively ( 18. 34 ± 0. 94) % , ( 10. 89 ± 0. 77 ) % and ( 11.90 ± 1.33 ) %, which were all remarkably higher than those in the blank control group( 2.45 ± 0. 09 ) % ( P 〈 0.05 ) . The cell apoptosis rate of 85℃ group was significantly higher than that of 70℃ or 55℃ group ( P 〈 0. 05). Conclusion In vitro, RFA may cause progressive injury to tumor ceils, and induce apopotosis of the residual tumor cells around the focus of RFA.
