摘要
目的探讨在不同处理条件下生成的钛酸钙(CaTiO3)涂覆的纯钛片,对成骨细胞生长的影响。方法利用不同条件下水热合成法制备CaTiO3涂覆的纯钛片,通过扫描电镜(SEM)、X射线衍射(XRD)对涂覆前后的钛片进行分析;将成骨细胞与材料共培养,利用显微镜观察法分别观察成骨细胞在不同处理材料表面的铺展状态,采用考马斯亮蓝法测定相对蛋白质含量、碱性磷酸酶(ALP)活性,并进行统计学分析。结果 CaTiO3涂覆的纯钛片的相对蛋白质含量和ALP活性,与对照组(无CaTiO3涂覆的纯钛片)比较,差异有统计学意义(P<0.05,P<0.01)。但CaTiO3涂覆的纯钛片在120、150℃和不同时间处理后对成骨细胞ALP活性及相对蛋白质含量未见明显改变。结论成骨细胞离体条件下,在120、150℃和不同时间处理后CaTiO3涂覆的纯钛片对成骨细胞的增殖和分化有促进作用,对ALP活性和相对蛋白质含量具有增高的影响,CaTiO3涂覆的纯钛片表面更有利于成骨细胞的黏附、增殖且对成骨细胞无毒性作用。
Abstract Objective To evaluate the impact of Calcium Titanate ( CaTiO3 ) coated pure titanium on the growth of osteoblasts. Methods The uniform CaTiO3 coatings on Ti substrates, prepared by a simple hydrothermal synthesis method, which was characterized by means of SEM and XRD before and after the modification. Spreadability status of osteoblasts on the surface of the different treatment materials was observed with microscope. The determination of relative protein content (Coomassie blue), alkaline phosphatase (ALP) activity of osteoblasts was measured by coculture with materials and analysized statistically. Results The protein content and alkaline phosphatase activity of CaTiO3 coated titanium piece at different temperature and time, as compared with the control group (no CaTiO3 coating of titanium), the difference had statistical significance (P 〈 0.05, P 〈 0. 01 ). CaTiO3 coating titanium sheet processed at 120, 150 ℃ temperature and different time had no significant changes on the ALP activity and relative protein content. Conclusion The CaTiO3 coated titanium piece, after processed at 120, 150 ℃ for different time, has a promotion effect on the proliferation and differentiation of osteoblasts under isolated conditions and improved ALP activity and relative protein content. Calcium Titanate coated titanium sheet surface is more conducive to osteoblasts adhesion, proliferation without toxic effects on osteoblasts.
