香菇多糖磷酸化修饰的工艺研究 被引量:12

摘要 利用多糖结构修饰原理进行香菇多糖磷酸化的研究,探讨其磷酸化修饰的最佳工艺条件。以磷酸化香菇多糖的磷酸根接枝量和黏度为检测指标,通过单因素试验考察磷酸化试剂用量、温度、时间和pH值对香菇多糖磷酸化修饰结果的影响,优选工艺条件,进而通过响应面设计法优化获得香菇多糖磷酸化修饰的最佳工艺条件为:三聚磷酸钠、三偏磷酸钠浓度分别为49.8 g·L-1、20.2 g·L-1,反应温度为81℃,反应时间为5.2 h,反应pH值为8.0,在该反应条件下,所得的磷酸根接枝量为8.99%。其中反应温度和反应pH值对香菇多糖磷酸化程度影响较为显著。 The conditions of lentinan phosphorylation were investigated according to the principle of polysaccharide structure modification. With the graft quantity of phosphate and viscosity as the detection index, the single factor experiments with the factors including phosphorylation reagent, temperature, time and pH values, were used to ex- plore their effects on lentinan phosphorylation. The response surface methodology was further used to optimize the lentinan phosphorylation conditions. The experimental results showed that the optimum conditions were obtained as 49.8 g ·L^-1 sodium tripolyphosphate, 20.2 g ·L^-1 sodium trimetaphosophate, 81 ℃ reaction temperature, 5.2 h reaction time and pH = 8.0. Under these reaction conditions, the phosphate graft quantity reached 8.99%, and the effects of reaction temperature and pH on the lentinan phosphorylation were remarkable.
