摘要 目的:采用分子杂交技术观察补阳还五汤对Aβ1-40所致老年痴呆(AD)大鼠海马区β-APP mRNA、IL-1βmRNA、IL-6 mRNA及TNF-αmRNA表达的影响,进一步探讨该组方治疗AD的机制,为其临床应用提供实验依据。方法:70只Wistar雄性大鼠随机分为正常组、假手术组、模型组、补阳还五汤组、补气组、活血组和脑复康组,每10只。采用注射Aβ1-40的方法制备AD模型,分子杂交技术检测大鼠海马区β-APP mRNA、IL-1βmRNA、IL-6 mRNA、TNF-αmRNA的表达。结果与结论:补阳还五汤能够明显降低AD大鼠海马区β-APP mRNA、IL-1βmRNA、IL-6mRNA和TNF-αmRNA的表达,从而改善大脑记忆障碍,达到防治AD的作用。 In this study,molecular hybridization technique is used to observe the effect of Buyang Huanwu decoction on the expression of β-APP mRNA,IL-1β mRNA,IL-6 mRNA and TNF-α mRNA of hippocampus of Alzheimer Disease(AD)rats induced by Aβ1-40,to further explore the mechanism of the treatment of AD,and to provide experimental evidence for the clinical application.The results show that Buyang Huanwu decoction can significantly reduce the β-APP mRNA,IL-1β mRNA,IL-6 mRNA and TNF-α mRNA expression in AD hippocampus,thus to improve memory disorders and to play a role in the prevention of AD.
