摘要
目的:探讨雷公藤甲素对人子宫内膜癌细胞株HEC-1B侵袭能力的影响。方法:雷公藤甲素干预HEC-1B细胞后,以明胶酶谱法检测细胞侵袭相关蛋白MMP-2/MMP-9的表达;Transwell法检测细胞侵袭能力的改变。结果:随着雷公藤甲素作用时间的延长,侵袭相关蛋白MMP-2/MMP-9的表达逐渐降低,HEC-1B细胞的侵袭能力亦逐渐下降,呈时间依赖性。结论:雷公藤甲素能通过下调侵袭相关蛋白MMP-2/MMP-9的表达,从而抑制HEC-1B细胞的侵袭能力。
Objective:To investigate the effect of triptolide on the invasion of human endometrial carcinoma cell line (HEC-1B). Methods:Gelatin zymography was performed to investigate the effects of triptolide on the activity of the invasion-related gelatinase proteins MMP-2 and MMP-9 in HEC-1B cells. Transwell was employed to assess the influence of triptolide on the invasion of HEC-1B cells. Results:The activity of the invasion-related gelatinase proteins MMP-2 and MMP-9 and the invasion of HEC-1B cells were decreased in a time-dependent manner,when exposed to triptolide. Conclusion:Triptolide could inhibit the invasion of HEC-1B by down-regulating the activity of the invasion-related gelatinase proteins MMP-2 and MMP-9.
