摘要
目的观察不同给药途径及不同给药时间下,血必净注射液对脓毒症大鼠血清炎症介质的影响。方法将91只健康Sprague-Dawley大鼠随机分为正常对照组(n=7)、脓毒症组(n=21)、灌胃给药+脓毒症组(预处理组,n=21)、静脉给药+脓毒症组(预处理组,n=21)和脓毒症+静脉给药组(治疗组,n=21)。除正常对照组外,其余各组按手术后3、6、12 h再分3组,每组7只。采用盲肠结扎穿孔法制作脓毒症模型。给药各组分别于术前或术后通过灌胃或静脉不同途径给予血必净(5 ml/kg)。各组于不同时间点留取血清标本测定肿瘤坏死因子-α(TNF-α)、一氧化氮(NO)、内皮素-1(ET-1)含量。结果预处理组及治疗组大鼠血清炎症介质含量较脓毒症组减低,预处理组减低更明显,差异有统计学意义(P<0.05);预处理组中,静脉给药较灌胃给药对脓毒症大鼠血清炎症介质的抑制作用更明显,差异有统计学意义(P<0.05)。结论血必净能抑制脓毒症大鼠血清TNF-α、NO、ET-1的过度生成与释放,预先静脉给药效果更佳。
Objective To observe the effect of injecting Xuebijing via different routes, as either treatment or pretreatment, on the changes of levels of inflammation mediators in serum in a rat model of sepsis. Methods Ninety-one healthy Sprague-Dawley rats were randomly divided into five groups: (1)control group, (2)sepsis group, (3)intragastric pretreatment group, (4)intravenous pretreatment group, (5) intravenous treatment group. Sepsis was induced by cecal ligation and puncture (CLP). For the pretreatment groups, Xuebijing was administered either via intragastricly or intravenously 2 hours before CLP at a dose of 5 ml/kg. For the treatment animals, Xuebijing was intravenously infused 2 hours after CLP at a dose of 5 mL/kg. The levels of tumor necrosis factor-α (TNF-α), nitric oxide (NO), and endothelin- 1 (ET- 1 ) in serum were detected at the different observation time. Results The levels of inflammation mediators in serum were significantly decreased in pretreatment and treatment groups compared with the untreated sepsis group, more obviously in pretreatment group (P〈0.05). The levels of inflammation mediators were significantly decreased in the intravenous pretreatment group compared with the intragastric pretreatment group (P〈0.05). Conclusion Irrespective of the route, the administration of Xuebijing significant decreased the levels of inflammation mediators in serum and intravenous pretreatment exerted the most significant depressive effect in this rat model of sepsis.
