摘要
目的观察外周血中表面活性蛋白A(SP-A)含量及其基因表达与慢性阻塞性肺疾病(COPD)的关系。方法按照COPD患者的肺功能第1秒用力呼出容积占用力肺活量之比(FVE1%)分级标准,选取稳定期Ⅰ、Ⅱ、Ⅲ、Ⅳ级非吸烟患者各15例,健康非吸烟对照组30例,分别测定血清中SP-A水平及其基因表达情况。结果非吸烟COPD患者血清中SP-A水平均高于健康非吸烟对照组,(55.38±2.59)μg/L、(68.36±2.50)μg/L、(76.55±2.12)μg/L、(96.20±4.44)μg/L vs(43.83±1.70)μg/L(P<0.01),血清中SP-A水平与FEV1%呈负相关(r=-0.881,P<0.01),SP-A基因在COPD非吸烟患者中表达高于健康非吸烟对照组(P<0.05),并与FEV1%呈负相关(r=-0.651,P<0.01)。结论外周血SP-A含量及其基因表达高低与稳定期COPD的病情严重程度有关。
Objective To observe the peripheral blood content of surfactant protein A(SP-A) and its gene expression for their relationship with chronic obstructive pulmonary disease(COPD). Methods In accordance with the lung function FVE1% grading standards of the COPD patients,the study included the stable phase I , II , III , IV-class non-smoking patients, respectively 15 cases, 30 subjects of healthy non-smoking group, serum SP-A content and its gene expression were measured respectively by ELISA and RT-PCR. Results SP-A contents in the serum of non-smoking patients with COPD stable phase I , II, III, IV-class were significantly higher than that of healthy non-smoking control group,(55.38±2.59) μg/L, (68. 36dz2. 50) μg/L,(76. 55±_2.12) rig/L,(96.20i4.44) μg/L vs (43.83±1.70) μg/ L( P 〈0.01) ,and a negative correlation with FEV1 % existed( r = -0. 881, P 〈0.01) ,SP-A gene in patients with COPD non-smoking patients expressed higher than that in healthy non-smoking control group ( P 〈0.05), and was negatively correlated with FEV1 % ( r = -0. 651, P 〈0.01). Conclusion The peripheral blood content of SP-A and its gene expression correlated with the severity of stable COPD.
