摘要
目的研究乙烯利对小鼠外周血淋巴细胞周期的影响。方法将24只健康成年SPF级雄性昆明小鼠随机分为4组,分别为阴性对照(0 mg/kg)组和低(1/32 LD50,85 mg/kg)、中(1/16 LD50,170 mg/kg)和高(1/8 LD50,340 mg/kg)剂量组,每组6只。灌胃染毒,每天1次,连续染毒14 d。测定小鼠外周血血液学指标和淋巴细胞周期的构成情况。结果各剂量乙烯利染毒组小鼠外周血小鼠红细胞计数(RBC)、白细胞计数(WBC)、淋巴细胞计数(LYM)、血小板计数(PLT)和血红蛋白(HGB)、嗜中性粒细胞(NEU)含量及淋巴细胞百分比(LYM%)以及淋巴细胞G0/G1期、S期、G2/M期细胞所占比例与阴性对照组相比,差异均无统计学意义。结论本研究未发现乙烯利引起小鼠外周血血液学各指标的改变,对淋巴细胞细胞周期分布无明显影响。
Objective To study the effect of ethephon on distribution of cell cycle of peripheral blood lymphocyte of mice by flow cytometry (FCM). Methods Twenty-four SPF health adult Kunming male mice were randomly divided into four groups, control (0 mg/kg body weight) ,low (85 mg/kg body weight) ,moderate (170 mg/kg body weight) and high (340 mg/kg body weight) dose groups. The mice were treated with ethephon by gavage (20 ml/kg),onee a day for 14 consecutive days. The blood was collected 28 days after the first gavage. The haematologieal indexes and cell cycle of peripheral blood lymphocyte in mice were examined. Results Compared with the control group, as increasing of ethephon dose, the levesl of mouse red blood cell count (RBC), white blood eel1 count (WBC), lymphocyte count (LYM), platelet count (PLT) and hemoglobin (HGB), neutrophil (NEU) content and percentage of lymphoeytes (LYM%) were not changed, and the cell cycle progression of mice peripheral blood lymphocyte was not blocked. Conclusion Ethephon may not affect the haematological indexes and cell cycle of peripheral blood lymphocyte in mice obviously. Key words: Ethrel ; Lymphocytes ; Cell cycle ; Blood
