丙烯腈对大鼠内皮祖细胞CD133表达和增殖功能的影响

摘要 目的探讨大鼠慢性丙烯腈(acrylonitrile,ACN)染毒模型外周血内皮祖细胞(endothelial progenitor cells,EPCs)增殖活性和CD133表达的变化。方法将30只健康3月龄雄性SD大鼠随机分为3组,分别为对照(蒸馏水)组和低(25mg/kg,1/4 LD50)、高剂量(50mg/kg,1/2 LD50)丙烯腈染毒组,每组10只。采用灌胃方式进行染毒,染毒容量为5ml/kg,每天1次,连续染毒3个月。采集大鼠外周血,分离培养EPCs。采用流式细胞仪检测细胞CD133的表达,采用MTT比色法测定细胞的增殖活性。结果对照组和低、高剂量丙烯腈染毒组EPCs中CD133的阳性表达率间比较,差异无统计学意义。第3-9天,与对照组比较,各剂量丙烯腈染毒组EPCs的增殖活性均下降,差异有统计学意义(P〈0.05);高剂量丙烯腈染毒组EPCs的增殖活性均低于低剂量丙烯腈染毒组,差异有统计学意义(P〈0.05)。结论丙烯腈对血管内皮功能的损伤作用可能与其抑制EPCs的增殖活性有关。 Objective To investigate the effects of acrylonitrile on CD133 and proliferation of rat endothelial progenitor cells from peripheral blood. Methods Thirty male rats aged three months were randomly divided into three groups,10 rats in each, the control group (normal saline ) ,low dose of aerylonitrile group (25 mg/kg body weight, 1/4 LDso) and high dose of aerylonitrile group(50 mg/kg body weight, 1/2 LDs0).The exposure was conducted through garage. Exposed capacity was 5 ml/kg, once daily, for three consecutive months. EPCs were isolated and cultured from peripheral blood of rat. CD133 expression were detected by flow cytometry, cell proliferation was assessed by MTT colorimetry. Results No significant difference was seen in CD133 positive expression rates among three groups. From the 3rd to 9th day of exposure, compared with the control group, the proliferation activity of EPCs in the acrylonitrile groups were all significantly declined (P〈0.05). The proliferation activity of EPCs in higher dose group was lower than that of lower dose group (P〈0.05). Conclusion Acrylonitrile may damage the vascular endothelium through the inhibitory effect on the proliferation activity of EPCs.
