紫苏愈伤组织诱导及继代培养条件优化 被引量:2

摘要 对紫苏愈伤组织诱导培养基、外植体、激素、光照条件以及基因型进行筛选,对愈伤组织继代培养中接种量和培养时间对增长量的影响进行探讨。结果表明,MS+6-BA 3.0mg/L+NAA 0.3mg/L诱导紫苏下胚轴愈伤组织效果最好,而MS+6-BA 2.0mg/L+2,4-D 1.0mg/L诱导子叶愈伤组织效果最好,光照条件为12h/d时有利于愈伤组织的诱导。供试10个不同基因型紫苏中,ZB-1愈伤诱导率最高。接种密度1.2g/皿(直径10cm培养皿),培养时间24d紫苏愈伤组织继代培养增长量高,愈伤品质好。 In this research, suitable medium, screened for perilla callus induction, and the explant, hormone, light conditions and genotypes were effects of inoculating quantity and subculture time on the fresh mass increasment of callus were studied. The results showed that it was optimal for MS+6-BA 3.0 mg/L+NAA 0.3 mg/L to inducing hypocotyl callus and MS+6-BA 2.0 mg/L+2, 4 D 1.0 mg/L to cotyledon callus, respectively. 12 h/d light condition was conducive to the growth of perilla callus. Among 10 perilla genotypes in this study, ZB-1 was the most suitable for the callus induction. The subcultured perilla callus grew faster and the quality was better under the conditions of seeding density 1.2 g/dish (10 cm diameter petri dish) and culture time 24 d.
