摘要
采用正交试验法 ,使里氏木霉 (Trichodermareesei)DWC 5在 50℃ ,转速 1 1 0r·min-1的摇瓶中发酵 5~ 7d。研究了铁、锰、锌、钴 4种微量元素对里氏木霉DWC 5纤维素酶产生的影响。结果表明 :对CMC酶活、FP酶活影响显著性的主次顺序依次为铁 >锌 >钴 >锰。微量元素的最佳组合对于CMC酶活 :Fe,Zn ,Co,Mn的用量分别为 5 .0 ,4.5 ,3 .8,3.8,4.0mg·L-1 ,对于FP酶活 :Fe,Zn,Co,Mn用量分别为 5 .0 ,4.5 ,3.8,3 .8mg·L-1 。此条件下CMC酶活、FP酶活分别为 1 32mgG/ml·30min,8.
Effect of four kinds of trace elements on cellulase activity from Trichoderma reesei, incubating on a rotary shaker, 110 r·min -1 ,5~7 days at 50 ℃,was investigated by using orthogonal test. The results showed that the effect factors on enzyme activity, in order of preference, were Fe, Zn, Co, Mn. Optimum condition of trace elements was Fe 5.0 mg·L -1 , Mn 4.5 mg·L -1 , Zn 3.8 mg·L -1 , Co 4.0 or 3.8 mg·L -1 for CMC enzyme activity, and that was Fe 5.0 mg·L -1 , Mn 4.5 mg·L -1 , Zn 3.8 mg·L -1 , Co 3.8 mg·L -1 for FP enzyme activity.Under those conditions, the enzyme activities towards CMC and filter paper reached 132 mg glucose/ml(supernatant)·30min and 8.4 mg glucose/ml(supernatant)·h.
