摘要
应用分子生物学技术构建了LacZ报告基因的pcDLacZ真核表达载体 ,并对多种促进基因表达的物质进行了筛选。应用筛选的促进基因表达物质对pcDST基因疫苗的免疫增强效果进行了观察。结果 :构建的LacZ报告基因的pcDLacZ真核表达载体 ,肌注后 1天就有表达 ,到第 7天达到高峰。通过对多种促进基因表达的物质进行筛选证明 ,4.5 %斯苯、1 %甘油、2 5 %蔗糖有明显促进基因的表达作用。 4.5 %斯苯、1 %甘油能促进pcDST猪瘟基因疫苗的免疫效果。表明 :4.5 %斯苯、1
The eukaryotic expression plasmid, pcDLacZ, was constructed and several kinds of compound which improve the expression and immune response of DNA vaccines in vivo were screened. The results demonstrated that The eukaryotic expression plasmid, pcDLacZ, was constructed by inserting LacZ gene into pcDNA3 as a report gene; 4.5% span and 1% glycerol could improve expression and immune response in vivo by DNA vaccines. 4.5% span and 1% glycerol can be used as adjuvant to enhance the immune response of DNA vaccine
