摘要
目的观察给以不同剂量氟的大鼠胰岛素水平的变化。方法经灌胃给氟复制氟中毒模型。利用Siemens ADVIA Centaur XP全自动化学发光免疫分析仪检测大鼠血清胰岛素水平,酶联免疫吸附法分析糖化血红蛋白水平,使用贝克曼全自动生化分析仪检测血脂。结果胰岛素和糖化血红蛋白水平的变化显示出低剂量和高剂量的氟都能够诱导胰岛素分泌,其高剂量氟组大鼠胰岛素水平更高。高剂量氟对胰岛素耐受影响不大,而低剂氟组大鼠胰岛素耐受能力下降,联合胰岛功能受损,进一步降低了大鼠对胰岛素的敏感性。结论一定剂量的氟刺激大鼠胰岛素的分泌,且对胰岛素的分泌成剂量-效应关系。此外,氟能够影响大鼠对胰岛素的敏感性,甚至降低其敏感性。
Objective To study the effect of fluoride on insulin level of rat.Methods Different doses of fluoride were administrated to rats through gavage.ADVIA Centaur XP Immunoassay(Siemens Healthcare Diagnostics) was used to test serum insulin level,and HbAlC was tested by ELISA technique.Blood lipid was detected by Beckman automatic biochemistry analyzer.Results It showed that low and high dose of fluoride induced the secretion of insulin,and higher fluoride administration caused higher level of serum insulin.High dose of fluoride affected little on insulin sensitivity,but low dose of fluoride reduced the insulin sensitivity of rats.Conclusion Certain dose of fluoride stimulated the insulin secretion,and showed the dose-response relationship.On the other hand,fluoride affected the insulin sensitivity and even reduced it.
