AQP7表达水平对砷染毒小鼠心脏组织砷含量的影响

Impact of AQP7 expression level on arsenic content in cardiac tissue of mice

摘要 目的了解AQP7表达水平的改变是否会引起砷(As3+)染毒小鼠心脏组织中砷含量的改变。方法雄性ICR小鼠48只,随机分为4组:胰岛素+砷组和生理盐水+砷组;匹格列酮+砷组和玉米油+砷组,每组12只。胰岛素+砷组以腹腔注射的方式给予胰岛素0.30~0.5 IU/d,其对照组给予等体积的生理盐水。匹格列酮+砷组以腹腔注射的方式给予匹格列酮4 mg/kg·d,其对照组给予等体积的玉米油。4周后,尾静脉注射0.4 mg/kg As2O3,15 min后取小鼠心脏组织,原子荧光分光光度法检测其砷含量,比较各组砷含量的差异。半定量RT-PCR检测AQP7表达水平的变化。结果(1)胰岛素+砷组心脏组织中砷含量(3517.37±562.29)ng/g与其对照组(4164.26±289.52)ng/g相比显著降低(P〈0.05)。半定量RT-PCR结果,胰岛素+砷组心脏组织中AQP7的表达水平与其对照组相比显著降低(P=0.009)。(2)匹格列酮+砷组心脏组织中砷含量(4249.20±748.22)ng/g与其对照组(4065.72±402.22)ng/g相比略有升高,但差异无统计学意义(P=0.651)。半定量RT-PCR结果,匹格列酮+砷组心脏组织中AQP7的表达水平增加,但与其对照组相比差异无统计学意义(P=0.053)。结论三价无机砷可能是通过细胞膜上水甘油通道蛋白AQP7进入心脏组织。 Objective To investigate if the change of the expression level of AQP7 would have an impact on the arsenic content in cardiac tissue of mice when they were exposed to arsenite.Methods Forty-eight male ICR mice were randomly divided into 4 groups: insulin and arsenic group,saline and arsenic group,pioglitazone and arsenic group and corn oil and arsenic group,with 12 mice in each group.The mice in insulin and arsenic group were given insulin via intraperitoneal injection(0.30 ~ 0.5) IU / d,and those in the control group were given an equal volume of saline.The mice in pioglitazone and arsenic group were given pioglitazone by intraperitoneal injection of 4mg / kg / d,and those in the control group were given the same volume of corn oil.After 4 weeks,all mice were given 0.4mg / kg arsenic trioxide by tail intravenous injection.The mice sacrificed 15 minutes later,and then the heart tissues were taken out for arsenic detection.Arsenic contents were determined by atomic fluorescence spectrum.The expression level of AQP7 was detected by reverse transcription-polymerase chain reaction(RT-PCR).Results The arsenic content in cardiac tissue significantly decreased in insulin and arsenic group {(3517.37 ± 562.29) ng / g} when compare with those in the control group {(4164.26 ± 289.52) ng / g}.The results of RT-PCR showed that the expression level of AQP7 in cardiac tissue significantly decreased in insulin and arsenic group(P = 0.009);The arsenic content in cardiac tissue increased in pioglitazone and arsenic group {(4249.20 ± 748.22) ng / g} when compare with those in the control group {(4065.72 ± 402.22) ng / g},however,the difference was not statistically significant(P = 0.651).The expression level of AQP7 in cardiac tissue in pioglitazone and arsenic group slightly increased,and the difference was not statistically significant(P = 0.053).Conclusion Trivalent arsenic may was transported into cardiac tissue of mice by AQP7.
