摘要
目的研究过敏性休克死亡豚鼠心、气管、喉头白细胞介素10(Interleukin-10,IL-10)表达的死后变化规律,寻找过敏性休克死亡的法医学诊断的客观指标。方法参照文献建立豚鼠过敏性休克死亡动物模型,采用免疫组织化学的方法,应用图像分析技术,检测各组豚鼠心、气管、喉头IL-10在死后0h、48h(4℃冷藏)、5d(-20℃冷冻)、7d(-20℃冷冻)四个时间点的表达量。结果在健康的豚鼠(对照组)心肌细胞、气管、喉头粘膜上皮细胞中IL-10均呈弱阳性表达,而在过敏性休克死亡豚鼠(实验组)心肌细胞、气管、喉头粘膜上皮细胞中IL-10均呈较强阳性表达,各实验组与相应的对照组之间均有显著差异(p<0.05);随着死后时间的延长,在死后冷藏48 h或冷冻7 d内,IL-10在心、气管、喉头的表达量呈缓慢减少趋势,但各实验组之间IL-10的表达量均无显著差异(p>0.05)。结论死后冷藏48h或冷冻7d内尸体,其心肌细胞、气管、喉头粘膜上皮细胞IL-10呈较强阳性表达,这可能成为过敏性休克死亡法医学鉴定的较好的客观指标。
Objective To research the postmortem changes of the expressions of IL-10 in the hearts, tracheas and larynxes of guinea pigs died of anaphylactic shock and to seek the objective, diagnostic indexes in identifying it. Methods According to the documents, the anaphylactic shock model of guinea pigs was established. The myocardia, epitheliums of tracheas and larynxes of the guinea pigs were examined at Oh ,48h (kept at 4℃ ) ,5d (kept at -20℃ ) ,7d (kept at -20℃ ) after death respectively for the expressions of IL-10 by immunohistochemical methods and technical image analysis. Results In the myocardia, epitheliums of tracheas and larynxes of the guinea pigs, in contrast to the weak positive expressions of IL-10 in the healthy guinea pigs (the control groups), there were relatively strong positive expressions in the guinea pigs died of anaphylactic shock( the experimental groups). Significant differences between the experimental group and the corresponding control group were observed (each P 〈 0.05 ) as well. The expressions of IL-10 in the sequence of Oh, 48h, 5d, 7d, declined slightly. There was, thus, no significant difference among each experimental group ( each P 〉 0.05 ). Conclusion There were relatively strong positive expressions of IL-10 in the myocardia, epitheliums of tracheas and larynxes of the guinea pigs, which was kept within 48h (4℃) or seven days( -20℃ ). This may be hopeful to provide the pathological morphological evidences and become the objective index in the forensic practice of anaphylactic shock.
