摘要
目的:建立麻黄总多糖及均多糖ESP-B4的含量测定方法,为有效的控制麻黄多糖质量提供科学有效依据。方法:采用可见分光光度法测定麻黄总多糖含量,检测波长:490nm。采用HPLC-ELSD法测定均多糖ESP-B4的含量,使用日本ShodexTMsugar KS-805+KS-G色谱柱,流动相:超纯水,柱温25℃,流速0.5mL/min,蒸发光检测器参数为漂移管温度116℃,载气流速3.3L/min。结果:麻黄总多糖及均多糖ESP-B4分别在6.25~100μg(r=0.999 8,n=5),2~12μg(r=0.999 8,n=6)范围内具有良好的线性关系;平均回收率(n=6)分别为101.68%(RSD=1.73%),99.655%(RSD=2.9%)。结论:本实验采用的方法简便,精密度高,结果准确可靠,为麻黄总多糖中均多糖ESP-B4含量测定提供了一个新的定量方法,同时为麻黄药材的质量控制提供了科学可靠的参考依据。
Objective:To develop a method for determination of total polysaccharides and homopolysaccharides ESP-B4 in Ephedra sinica Stapf (Mahuang).Methods:Total polysaccharides were determined by visible spectrophotometry,and the detected wavelength was set at 490nm.The separation of homopolysaccharides ESP-B4 was detection by HPLC ELSD on the ShodexTM sugar KS-805 and KS-G column with water as mobile phases at the flow rate of 0.5mL/min; the column temperature was 25 ℃ ; the parameter of Evaporative Light-Scattering Detector (ELSD) was:Tube Temp:116℃,Gas Flow:3.3L/min.Results:The standard curves for total polysaccharides,homopolysaccharides ESP-B4 were linear in the ranges of 6.25 ~ 100μg(r =0.999 8,n =5) and 2 ~ 12 μg (r =0.999 8,n =6) ; the average recoveries (n =6) was 101.68 % (RSD =1.73 %),99.655 % (RSD =2.9 %),respectively.Conclusion:The method is simple,accurate and reliable.It will provide an new excellent quantitative method and reference for quality control of ephedra.
