5个温郁金居群遗传多样性的RAPD分析 被引量:4

摘要 采用随机扩增多态DNA(RAPD)分子标记技术对温郁金(Curcuma wenyujin)居群的遗传多样性进行分析.从瑞安等温郁金种植地中选取了5个居群50个个体进行RAPD-PCR扩增,共得到1551条扩增带和56个清晰而且可重复的有效结合位点,其中多态位点有54个,多态位点百分率为96.43%.5个居群内总的基因多样度平均值为0.3715,居群间的基因多样度为0.1904,居群内的基因多样度为0.1811,平均多态位点百分率为46.07%。基因分化系数平均值为0.5124,其中居群间的遗传变异占总遗传变异的51.24%,居群内的遗传变异占总遗传变异的48.67%.Nei's基因多样性指数和Shannon多态性信息指数分别为0.371 5和0.546 9,表明5个居群的总体遗传多样性处于较高的水平. The genetic diversity of populations of Curcuma wenyujin,an endemic species in Wenzhou(Ruian,Pingyang and Yueqing),are analyzed by the Random Amplified Polymorphic DNA(RAPD) approach.A total of 1 551 bands and 56 distinct and reproducible loci are obtained from 5 populations by 6 primers screened from a total of 40 primers.RAPD data shows that there are 54 polymorphic loci with a percentage of 96.4,implying a relatively high level of intraspecific genetic diversity.This investigation additionally indicates a high genetic polymorphism level within the populations of C.wenyujin with H:(0.371 5) and I:(0.546 9),and PPL(46.07%) within 5 populations.
