特发性血小板减少性紫癜患者外周血PBMC中颗粒溶素的表达及其意义

Expression of Granulysin in the Peripheral Blood Mononuclear Cells from Patients with Idiopathic Thrombocytopenic Purpura

目的 探讨特发性血小板减少性紫癜（ITP）患者外周血中颗粒溶素的表达及其意义.方法 收集确诊30例ITP患者、50例体检健康者全血,通过分离PBMC、抽提总RNA,采用MGB探针,以18s RNA为内参,经荧光定量PCR（FQ-PCR）方法,检测其颗粒溶素的mRNA表达水平.结果 ITP患者外周血PBMC中颗粒溶素的平均拷贝数（18.27±3.1）×109,显著高于体检健康者（3.94±0.95）×107（P〈0.05）.结论 颗粒溶素可能参与了ITP的发病过程.

Objective To investigate the expression and clinical significance of granulysin in the peripheral blood mononuclear cells （PBMCs） of idiopathic thrombocytopenic purpura （ITP） patients. Methods The granulysin mRNA relative expression in PBMCs from 30 ITP patients and 50 healthy individuals was determined by FQ-PCR methods. Results The granulysin mRNA level of ITP patients was higher than that of controls （P〈0. 05）. Conclusion Granulysin in PBMCs involved in the pathogenesis of ITP.
