化学诱导表达载体pKIGW在拟南芥中的应用

摘要 为了评价pKIGW载体在模式植物拟南芥中的可用性和稳定性,利用拟南芥转基因纯系植株,对其施加诱导物后,进行GUS染色并进行实时定量PCR试验。结果表明:GUS染色后发现转基因植株幼苗各器官均被染成蓝色,实时定量试验证明,目的基因在地塞米松诱导后,表达水平急剧上升。结论:该系统可精确控制目的基因的表达,基因功能分析的结果更可靠,是理想的基因功能鉴定表达载体。 The GUS staining and real-time quantitative PCR of induced transgenic Arabidopsis plants were conducted to evaluate the availability and stability of pKIGW vector in Arabidopsis. The results showed that all tissues of transgenic seedlings came into blue. Real-time PCR indicated that target gene expression increased sharply compared to those of control plants after transgenic seedlings were challenged by dexamethasone. These results demonstrated that the expressions of target gene and reporter gene can be well controlled by chemical inducer and pKIGW vector is an ideal expression system for precise characterization of gene function.
