不同包埋基质对青天葵人工种子萌发率的影响 被引量:4

摘要 【目的】以青天葵根状茎诱导的再生球茎作为种胚,考察不同包埋基质对青天葵人工种子萌发率的影响。【方法】采用6因素2水平的正交实验设计考察不同包埋基质:50.0 g/L海藻酸钠、1.0 mg/L 6-糠氨基嘌呤(KT)、0.5 mg/L青霉素、0.3 mg/L赤霉素(GA3)、0.5 mg/Lα-萘乙酸(NAA)、2.0 mg/L 6-苄基嘌呤(6-BA)对人工种子萌发率及生长的影响。【结果】50.0 g/L海藻酸钠+0.3 mg/L GA3+1.0 mg/L KT+0.5 mg/L青霉素的包埋基质组合对青天葵人工种子的萌发率达到85%以上,并且萌发比较整齐,萌发后长势良好。【结论】建立了青天葵人工种子的制作方法,获得了较高的萌发率。 Objective Using the corm regenerated from Nerviliafordii rhizomes as artificial seeds, the effect of different embedding matrixes on their germination rate was evaluated. Methods The germination rates of artificial Nervilia fordii seeds embedded in the various matrixes of sodium alginate (50.0g/L), kinetin (KT, 1.0 mg/L), penicillin (0.5mg/L), gibberellin(GA3, 0.3 mg/L), (α-naphthaleneacetic acid (NAA, 0.5 mg/L) and 6-benzylaminopurine (6-BA, 2.0 mg/L) were analyzed by the orthogonal test with 6-factor and 2-level design. Results The germination rate of artificial Nervilia fordii seeds embedded in the combination matrix of sodium alginate + GA3 + KT +penicillin was over 85%, and their spouts grew neatly and well. Conclusion A culture method for artificial Nerviliafordii seeds has been established, which brings in higher germination rate.
