摘要 目的探讨Toll样受体4(toll-like receptor 4,TLR4)在肾间质纤维化大鼠模型中的表达及意义;研究黑骨藤追风活络胶囊对肾间质纤维化的作用及可能的机制。方法采用单侧输尿管梗阻(UUO)致肾间质纤维化大鼠模型。将36只雄性SD大鼠随机分为假手术组、模型组、黑骨藤追风活络胶囊(HGT)治疗组(243mg/kg/d),手术后第7天、第14天分别处死各组中6只大鼠。用HE染色方法观察各组大鼠肾小管间质损伤程度,免疫组化方法观察各组大鼠肾组织中TLR4、转化生长因子-β1(transforming growth factor-β1,TGF-β1)的表达情况。结果模型组TLR4、TGF-β1的表达及肾小管间质损伤指数明显高于同期假手术组(P<0.01);黑骨藤追风活络胶囊治疗组TLR4、TGF-β1的表达及肾小管间质损伤指数明显低于模型组(P<0.01)。结论TLR4及其介导的信号通路可能在肾间质纤维化中起促进作用;黑骨藤追风活络胶囊对肾间质纤维化有一定的防治作用。 Objective To investigate the expression and significance of Toil-like receptor 4 (TLR4) in rat model of Tubulointerstitial fibrosis. To investigate the effect and mechanism of drug which were hei guteng zhui feng huo luo capsule (HGT) on the process of the fibrosis. Methods 36 normal male Sprague-Dawley rats were randomly divided into sham-operated controls,unilateral ureteral obstruction (UUO), HGT treatment (243 mg/d),6 rats of each group were sacrificed randomly on the 7 days, 14 days after surgery. The tubulointerstitial damage indexe of histological studies were implemented by HE. Immuno- histochemistry was performed on obstructed renal tissue to determine the expression of TLR4 and transforming growth factor-~ll (TGF-~I). Results The expressions of TLR4 and TGF-β1 and the tubulointerstitial damage indexs in UUO group at different time points were sig- nificantly higher than that in sham-operated group (P〈0. 01). The expressions of TLR4 and TGF-β1 in HGT group were significantly lower than that in UUO group (P〈〈0. 01) ,also the tubulointerstitial damage indexs. Conclusion TLR4 with its signal transduction progress may play a stimulative role in renal interstitial fibrosis. HGT can cure and prevent from renal interstitial fibrosis.
