甜瓜茎尖法遗传转化体系的建立 被引量:2

摘要 利用甜瓜材料"M-23"建立一种有效的、不依赖于组织培养的农杆菌介导甜瓜遗传转化体系,该遗传转化体系利用甜瓜植株茎尖组织作为受体材料,避免传统组织培养获得转化植株周期长、工作量大以及无菌操作要求高的缺陷,可在受体材料后代中直接筛选出转基因品系。研究优化了转化流程中的各项参数,并将CmACS-7基因连入pBI121载体对甜瓜进行遗传转化,对转基因后代种子的PCR检测和Southern杂交结果表明本转化体系转化率为8%。 An efficient and organ culture independent melon Agrobacterium-mediated genetic transformation protocol was developed by melon line M-23 in this study, this genetic transformation system used shoot apex tissue as Agrobacterium receptor tissue, thus avoiding the traditional tissue culture methods' limitations, such as long cycle, big workload and sterile operation. Transgenic plant could be gained directly in descendants of receptor materials by transgenic screening. This study optimized various parameters of the transformation process. And pBI121 vector containing CmACS-7 gene was transformed into melon material M-23, PCR detection and Southern hybridization of trans- genic offspring seeds results showed that the transformation frequency of this transformation system was 8%.
