三七、虫草菌丝对实验性肝纤维化预防作用的研究 被引量:7

摘要 目的 观察三七、人工虫草菌丝对CCl4中毒性肝纤维化的预防作用。方法 复制大鼠CCl4中毒性肝纤维化模型 ,并以秋水仙碱作为阳性治疗对照。分空白对照组、肝纤维化模型组、秋水仙碱组、三七组、虫草菌丝组。采用光镜、电镜观察肝脏组织学改变 ,测定血清ALT、白蛋白、球蛋白、HA、LN、肝组织Hyp含量 ,以反映肝细胞损伤及肝纤维化程度。将形态改变与功能变化相结合 ,对比分析虫草菌丝对CCl4中毒性肝纤维化的预防作用。结果 CCl4可引起肝组织严重脂肪变性及肝纤维化。各组肝组织损伤及肝纤维化的严重程度依次为 :模型组 >虫草菌丝组 >秋水仙碱组 >三七组。结论 本实验条件下 ,三七是较理想的预防肝纤维化的药物。虫草菌丝对肝脏CCl4损伤具有一定保护作用 ,有较强促进肝细胞再生修复作用 ,同时在损伤肝组织中成纤维细胞及胶原纤维也明显增生 ,这将加快肝纤维化进程 ,临床应用时应权衡利弊。秋水仙碱有一定预防肝纤维化作用 ,但毒副作用不可忽视。 Objective:To investigate the effects of Panax Notoginseng(PN)and Cordycesp Sinensis(CS)on CCl 4-induced hepatic fibrosis(HF).Methods:The model of CCl 4-induced HF in rats was treated with PN、CS or Colchicine (Col). The histological changes were observed with light and electronic microscopes. The Serum concentrations of alanine aminotransferase(ALT)、albumin、hyaluronic acid(HA)、Laminin(LN) were determined and the level of hydroxyproline(Hyp) in liver tissue was also measured in order to reflect the extent of hepatocytic lesion and HF. The Col group was set up as positive treatment control.Results:Severe fatty degeneration and HF can be induced by CCl 4 intoxication in rat model. The severe extent of hepatocytic lesion, fatty degeneration and infiltration of inflammatory cells was :Model group>CS group>Col group>PN group.Conclusion:PN is an ideal drug on anti chronic hepatic lesion and hepatic fibrosis. CS can exert significant effects on improving regeneration and damage of hepatocytes. Meanwhile, the proliferation of fibroblasts and collagenous fibers were also obviously observed. Col has certain effect on preventing hepatic fibrosis, but its side effect could not be neglected.
