摘要
目的 探讨S311抗癌菌苗能否增强腹水癌小鼠腹腔巨噬细胞和其他几种单核巨噬细胞的抗腹水癌作用 ,以及S311抗癌菌苗与IFN γ、IL 2在抗肿瘤增殖上是否具有协同作用 ,为S311抗癌菌苗免疫治疗胸腹水癌提供依据。方法 对腹水癌小鼠腹腔内注射S311抗癌菌苗、S311抗癌菌苗和细胞因子 ,连续 3周 ,分别于第 1、2、3周从小鼠体内分离腹腔巨噬细胞、脾巨噬细胞、肺巨噬细胞、血液单核细胞 ,并分别与鼠腹水癌细胞联合培养 ,用MTT法测定 4种效应细胞对腹水癌细胞增殖的抑制作用。结果腹腔注射S311抗癌菌苗可明显抑制艾氏腹水癌的生长 ,S311抗癌菌苗能明显增强 4种效应细胞抑制腹水癌细胞增殖 ,S311抗癌菌苗与IFN γ、IL 2存在着明显的协同作用 ,抑制率分别提高了 32 %~ 188%。
Objective To determine the mechanisms responsible for the effects of S311 anticancer vaccine on immunotherapy of peritoneal ascites cancer(PAC).Methods Mice with PAC received S311 anticancer vaccine alone or with IFN-γ and IL-2 intraperitoneally every other day for three weeks.The activated macrophages of every mouse were purified from abdominal cavity,spleen,lung blood,and cocultured with murine PAC at a ratio of 8 to 1.MTT assay was used to evaluate the effects of activated macrophages on the regression of PAC.Results The results showed S311 anticancer vaccine could significantly enhance cytotoxical effects of the macrophages on the regression of PAC.It also could be a cooperator with IFN-γ and IL-2.Combination therapy increased regression rate by from 32% to 188%.Conclusion Macrophages activated S311 anticancer vaccine may play an important role on immunotherapy of peritoneal ascites cancer.
