大鼠乳腺增生病模型的建立 被引量:6

摘要 目的建立大鼠乳腺增生疾病模型并观察其复旧情况。方法选用SD雌性未孕大鼠,随机分为对照组和模型组,模型组使用肌肉注射苯甲酸雌二醇(剂量为0.5 mg/kg·d)及黄体酮(剂量为5 mg/kg·d)方法复制乳腺增生模型,造模成功后取下右侧乳房,再去除造模因素饲养28 d后取下左侧乳房,分别与对照组对应乳房进行比较,观测第三对乳头直径、腺泡、导管数量以及腺泡横径和导管内径。结果模型组与对照组相比,乳头直径(pixel)、腺泡数量(个)、直径(pixel)、导管内径(pixel)均有增大(分别为:1.17±0.12 vs 0.93±0.097,23.75±5.30 vs13.13±3.61,360.13±65.63 vs 162.50±33.68,236.63±68.28 vs 102.13±54.04,P均<0.01);去造模因素组乳头直径(pixel)、腺泡数量(个)、横径(pixel)以及导管内径(pixel)均小于模型组,且差异有显著性意义(分别为0.96±0.10 vs 1.17±0.12,16.50±5.44 vs 23.75±5.30,228.50±44.10 vs 360.13±65.63及162.13±73.40 vs236.63±68.28,P均<0.01),各组间导管数量差异无显著性意义(P>0.05)。结论雌二醇、孕酮交替肌肉注射的方法可成功建立大鼠乳腺增生病模型,去除造模因素28 d后腺体复旧。SD大鼠乳腺增生病模型的稳定性有待进一步研究。 Objective To establish the model of hyperplasia of mammary gland in rat and observe the redintegration of the breast. Methods SD female non - pregnant rats were randomized into 2 groups: normal group and model group. Model group received muscular injection of (0.5 mg/kg · d) and progesterone (5 mg/kg · d) to induce hyperplasia of mammary glands. After the modeling, right breasts taken from all of rats, then the opposite breasts would be taken after 28 days with conventional feeding, both breasts of model group compared with the control group respectively by diameter of the third nip- ples, the quantity and diameter of acinar lumina and duct of mammary gland. Results Compare with control group, the di- ameter of the nipple, the quantity and dianleter of acinar lumina and ducts of mammary gland in model group increased significantly ( diameter of the nipple : 1.17 ± 0.12 vs 0.93 ± 0. 097, the quantity of acinar lumina : 23.75± 5.30 vs 13.13 ± 3.61 ,the diameter of aeinar lumina:360. 13± 65.63 vs 162.50±33.68, the diameter of duct: 236.63±68.28 vs 102.13 ±54.04. P 〈 0.01 ) , but compare with the model group, they reduce in remove the factors group ( diameter of the nipple : 0.96±0.10 vs 1.17± 0.12 ; the quantity of acinar lumina : 16.50 ± 5.44 vs 23.75 ±5.30 ; the diameter of acinar lumina : 228.50±44.10 vs 360.13 ± 65.63 ; the diameter of duct : 162.13 ± 73.40 vs 236.63±68.28, P 〈 0.01 ), there was no significant difference in quantity of ducts among groups ( P 〉 0.05 ). Conclusion A great quantity of ectogenic estradiol can induce mammoplasia, but without exogenous estrogen the model will be redintegrated after 28 days.
