摘要
目的建立快速有效标记肿瘤显像探针^11C-乙酸盐的简便方法,为临床诊断提供辅助用药。方法以^11CO2为原料,通过优化标记的原料CH,MgBr、水和10%H。PO。的用量,改变反应溶剂并采用在线固相萃取的方法,快速有效标记^11C-乙酸盐。同时以一例病理结果为肝癌的患者进行^18F—FDG和^11C-酸盐的PET/CT对比研究。结果标记^11C-乙酸盐的最佳标记条件为:2.0ml的CH,MgBr、0.2ml的水和0.6ml的10%的磷酸,用固相萃取小柱进行纯化得到^11C-乙酸盐,其校正产率达到(52.1±2.7)%,放射化学纯度为100%。PET/CT对比显像表明^11C-乙酸盐显像可以弥补^18F—FDG的不足,提高诊断的准确性。结论改进方法后标记^11C-乙酸盐的产率大幅提高,能满足临床的常规用药需求,^11C-乙酸盐临床PET/CT显像具有极大的应用价值。
Objective To establish a simple, rapid, valid method to label the tracerH C-acetate in an attempt to provide the adjuvant drug for clinical diagnosis. Methods ^11C-acetate was rapidly effectively labeled by using on-line solid phase extraction and optimizing the amounts of CH3 Mg- Br, water, and 10% H3PO4, with ^11C serving as raw material. A patient with liver cancer who was pathologically confirmed underwent 18F-FDG PET and 11C-acetate PET separately, and the results were compared. Results A satisfying yield of 11 C-acetate was obtained by solid phase extraction under optimal conditions (2. 0 mL CHaMgBr, 0. 6 mL 10% H3PO4, 0. 2 mL water) . The corrected production rate reached 52. 1 ±2.7% and the radiochemical purity 100%. PET/CT imaging showed that 11 C-acetate could compensate the shortage of 18F-FDG in imaging, and improve diagnostic accuracy. Conclusions The production of "C-acetate was significantly enhanced after modification of the method, which will meet the clinical need for routine use of the drug. "C-acetate PET imaging is of great clinical application value.
