乙醇沉淀操作对羚羊角塞注射液中肽类成分和游离氨基酸含量的影响

Effects of Ethanol precipitation Operation on the Content of Peptide Constituents and Free Amino Acid in Cornu Antelopis Blocks Injection

利用程序升气相色谱法、凝胶色谱法和阴、阳离子交换色谱法及UV光谱鉴测等方法研究了羚羊角塞注射液制备过程中 ,乙醇沉淀操作对其中的肽类和 18种游离氨基酸含量的影响。实验结果表明该操作使游离氨基酸总量损失 6 1% ,而总氨 (即肽类和氨基酸 )损失约 70 % ,阳离子交换色谱法和凝胶色谱法可分离纯化该注射液中的肽类成分 ,为乙醇沉淀工艺的使用和改进提供了实验依据。

The effects of ethanol precipitation operation on the content of peptide and eighteen kinds of free amino acids during the preparation of cornu antelopis blocks injection was studied with temperature programming gas chromatography、gel chromatography、anion-oand cation-exchange chromatography and ultra-violet spectroscopy.Results: indicated that this operation made the content of free amino acid lost 61%,and the total nitrogen(peptide and amino acid)lost about 70%,the piptides in this injection could be isolated and purified with cation-exchange chromatography and gel chromatography.These provided experimental basis for using and improving of ethanol precipitation process.
