摘要
为了探讨葛根提取物诱导白血病细胞凋亡的作用及机制。以 18例急性非淋巴细胞性白血病 (ANLL)患者骨髓及HL 6 0细胞分别与葛根提取物、高三尖杉酯碱 (HHT)、阿糖胞苷 (Ara -c)及HA(HHT +Ara c)共同培养 2 4h ,应用细胞形态学、DNA凝胶电泳、DNA片段百分率测定等方法观察以上处理对白血病细胞凋亡的影响 ;对其中 10例处理前后的骨髓细胞用免疫组化法进行bcl 2、c myc(B细胞淋巴瘤 /白血病 2、细胞 癌基因家族 )基因蛋白表达检测。结果 :处理各组均见典型凋亡形态和DNA梯状带 ,对照组无 ,葛根提取物处理后的骨髓细胞bcl 2、c myc基因蛋白表达明显下调。结论 :葛根提取物可诱导ANLL细胞和HL 6 0细胞发生凋亡 ;葛根提取物对bcl 2、c myc基因蛋白表达的下调或抑制可能是其促凋亡机制之一。
The purpose of the study was to investigate the effects and mechanism of pueralia Lobata (PL)on the apoptosis of leukemia cells.The target leukemia cell line HL-60 and bone marrow leukemia cells from 18 acute nonlymphocytic leukemia(ANLL)patients(3M ! 7M 2 6M 3 2M 5)at presentation were studied.Apoptosis was detected by morphological observation,fluorescence assay,DNA electrophoresis,Percentage of DNA fragmentation test.It was concluded that PL could induce apoptosis in ANLL cells and HL-60 cells.
