摘要
目的 探讨HSV tk GCV系统联合rhTNF α抗脑胶质瘤的有效性。方法 利用重组HSV tk逆转录病毒载体转染大鼠C6细胞得到HSV tk阳性细胞 ,C6 GCV、C6+tk GCV、C6+rhTNF α和C6+tk GCV +rhTNF α4组在体外分别给予GCV或 和rhTNF α处理 ,用MTT法测量细胞存活率 ,比较各组肿瘤细胞存活率。卡方检验作显著性检测。结果 未加任何抗癌因子处理的C6细胞组肿瘤细胞生长旺盛 ,细胞形态无明显改变 ;各处理组的肿瘤细胞 2~ 3天后开始逐渐出现细胞突触消失 ,细胞变圆 ,胞体皱缩 ,核仁固缩、裂解、甚至消失 ,继而逐渐死亡。联合应用HSV tk GCV +rhTN α组的肿瘤细胞存活率比单纯应用HSV tk GCV组或rhTNF α组明显低 (P <0 0 5)。结论 :联合应用HSV tk GCV系统和rhTNF α的杀癌细胞效果优于单一使用HSV tk GCV系统或者rhTNF α。
Objective To investigate the synergistic anti-glioma effects of HSV-tk/GCV system and Recombinant Human Tumor Necrosis Factor-alpha(rhTNF-α)in vitro.Methods The HSV-tk gene was transferred into C6 cells to obtain C6 tk + cells.The test was divided into four groups:C6 cells group、C6tk/GCV group、C6+rhTNF-α group and C6+tk/GCV+rhTNF-α group.Each group was treated with GCV or/and rhTNF-α.The cells survival rates of every group were determined by MTT technique.Results The tumor cells without any anti-glioma factor treatment were growing better and no change was observed in morphology.After 2-3 days,cell axons disappeared,cell bodies crimpled,nucleoli crinkled and cracked in each treated group.The cells survival rates in C6+tk/GCV+rhTNF-α group were lower than that in C6 tk/GCV group and C6+rhTNF-α group ( P <0.05).Conclusion The effect of combination therapy for glioma with HSV-tk/GCV system and rhTNF-α is better than either that of HSV-tk/GCV or rhTNF-α.
