摘要 目的:研究高表达缺氧诱导因子(HIF-1α)对肾癌(GRC-1)细胞增殖和细胞内斯钙素-1(STC-1)、Ca2+水平的影响。方法:构建HIF-1α重组质粒,转染GRC-1细胞,MTT法检测GRC-1细胞的增殖,RT-PCR法检测GRC-1细胞内HIF-1α及STC-1的表达,荧光探针检测细胞内Ca2+水平,同时设正常GRC-1细胞作为对照组。结果:与对照组相比,转染了HIF-1α的GRC-1细胞增殖率和STC-1表达水平显著增高(P<0.05),Ca2+水平明显降低(P<0.05)。结论:HIF-1α可能参与了肾癌细胞的恶性增殖,其机制可能与STC-1下调肿瘤细胞内Ca2+水平有关。 Objective: To investigate the effect of high expression of hypoxia inducible factor-lα (HIF- lα) on renal carcinoma (GRC-1) cell proliferation and the levels of stanniocalcin (STC-1) and Ca^2+ inside the cells. Methods: HIF-lα recombinant plasmid was constructed and transfected GRC-1 cells. The GRC-1 proliferation was observed with MrlT assay. RT-PCR was used to assess the expression levels of HIF-lα and STC-1. Ca^2+ levels'were tested by fluorescence probe. GRC-I cells that did not received transfection served as control group. Results:Proliferation rates and STC-1 levels in HIF-lα transfected GRC-1 cells were higher and Ca^2+ level was lower than those in GRC-1 of control group ( P 〈 0.05). Conclusion :HIF-lα is involved in malignant proliferation of renal carcinoma cells, The mechanism may relate to down-regulating of Ca^2+ level in the cell by STC-1 .
