摘要
目的:观察高迁移率族蛋白B1(HMGB 1)在肾小管间质纤维化大鼠模型中的表达,探讨肾炎舒对肾小管间质纤维化的疗效及作用机制。方法:将36只大鼠随机分为假手术组、模型组、肾炎舒治疗组[378 mg/(kg·d)],采用单侧输尿管结扎术(UUO)制作肾小管间质纤维化大鼠模型,术后第7天及第14天分别处死各组中6只大鼠。检测各组大鼠肾小管间质损害程度及组织中HMGB 1、转化生长因子-β1(TGF-β1)的表达。结果:模型组HMGB1、TGF-β1的表达及肾小管间质损伤指数明显高于同期假手术组(P<0.05),肾炎舒治疗组HMGB1、TGF-β1的表达及肾小管间质损伤指数明显低于同期模型组(P<0.05)。结论:HMGB1可能协同TGF-β1促进肾小管间质纤维化,肾炎舒胶囊可抑制肾组织中HMGB1及TGF-β1的表达,对肾间质纤维化有一定的防治作用。
Objective: To investigate the expression of high mobility group protein B1 (HMGB1) in rat model of renal tubule interstitial fibrosis (RIF), and to observe the curative effect of Shenyanshu (a traditional Chinese medicine) on renal interstitial fibrosis. Methods:Thirty-six normal male Sprague- Dawley rats were randomly divided into 3 groups : sham-operated control group ( group SC), RIF model group (group RM) and Shenyanshu treatment group (group ST), and RIF rat model was established by unilateral urethral obstruction (UUO). Rats of the 3 groups were sacrified on the 7^th day and 14^th day after surgery. The expression of HMGB1 and transforming growth factor-β1 (TGF-β1) in renal tissue and the extent of tubulointerstitial damage were detected by immunohistochemistry method. Resuits:The expression of HMGB1 and TGF-β1, and the tubulointerstitial damage indexes were significantly higher in group RM than in group SC ( P 〈 0. 05 ), and were significantly lower in group ST than in group RM ( P 〈0.05). Conclusion:HMGB1 can cooperate with TGF-β1 to accelerate renal interstitial fibrosis. Shenyanshu may inhibit the expression of HMGB1 and TGF-β1 in renal tissue, and in this way, it might have some preventive effect to renal interstitial fibrosis.
