摘要
目的:探讨甲状腺激素受体β1(thyroid hormone receptorβ1,TRβ1)和β连环蛋白(β-catenin)在胃癌组织中的表达及其临床意义。方法:应用免疫组化SP法和RT-PCR技术检测80例胃癌组织及40例无癌胃黏膜组织中TRβ1和β-catenin蛋白、mRNA的表达,分析两者mRNA表达量与临床病理指标的关系。结果:胃癌组织和无癌胃黏膜组织中TRβ1蛋白的阳性表达率分别为16.25%(13/80)、80.00%(32/40);β-catenin蛋白的异常表达率分别为62.50%(50/80)、0(0/40)。TRβ1 mRNA在无癌黏膜组织中的表达量高于癌组织(P<0.01),β-catenin mRNA则相反(P<0.01),且两者的表达量与肿瘤分化、淋巴结转移、浸润深度、TNM分期有关(P<0.05)。结论:胃癌组织中TRβ1蛋白和mRNA的表达均减少,β-catenin则增加;两者mRNA表达量与胃癌的生物学行为有关。TRβ1和β-catenin有可能参与胃癌的发生、发展和侵袭转移。
Objective:To explore the expression and clinical significance of TR β 1 and β-catenin in gastric cancer.Methods:Immunohistochemical SP method was used to detect the expression of TR β 1 and β-catenin protein,and RT-PCR was used to test their mRNA in 80 human gastric carcinomas and 40 noncancerous gastric tissues.Then,the correlation between the mRNA expression quantity and clinical indicators was analysed.Results:The positive expression rate of TRβ1 protein in gastric carcinomas and noncancerous gastric tissues was 16.25% (13/80) and 80.00% (32/40),respectively.The abnormal expression rate of β-catenin protein was 62.50% (50/80) and 0(0/40),respectively.TR β 1 mRNA expression quantity was higher in noncancerous gastric tissues than that in gastric carcinomas,whereas β-catenin mRNA presented an opposite tendency.(P<0.01).Both TRβ1 mRNA and β-catenin mRNA expression quantity were related with tumorous differentiation,lymphatic metastasis,infiltration and TNM stage.Conclusion:TRβ1 expression quantity is decreased,whether at the level of protein or mRNA,but β-catenin is increased at the both level.TR β 1 and β-catenin's mRNA quantity relate to tumorous biological behaviour.Therefor,TRβ1 and β-catenin probablely involve in gastric carcinoma's occurrence,development,aggression and metastasis.
