与乳酸菌具有共生作用酵母菌培养基的优化 被引量:6

摘要 以对乳酸菌(LC5)有促进作用的酵母菌(YE4)为研究对象,OD值和活菌数为测定指标,筛选合适的培养基。通过对5种酵母菌常用培养基的初筛及二次筛选,确定在YPD培养基中YE4生长良好。以YPD培养基作为基础培养基,进一步利用单因素试验和正交试验的设计方法对其进行优化,得出改良YPD培养基的最佳成分是大豆蛋白胨为0.4%,胰蛋白胨为0.4%,酵母浸提粉为0.4%,蔗糖为1.2%,蒸馏水为100 mL。改良YPD培养基活菌计数测得的活菌数可达到5.01×108CFU/mL,比YPD基础培养基高一个数量级;OD值是YPD基础培养基的1.06倍。 Yeast (YE4) is taken as research object, which can promote the rote of lactic acid bacteria (LCS) , and OD^0 and viable count are as determination of indicators to filter suitable medium of YE4. By selecting from 5 mediums which the YE4 usually used, the result shows that the YE4 grows very well in YPD medium. So YPD is selected as the base medium, by using single factor experiment and orthogonal test design method to optimize it, and the best ingredients of the modified YPD are that: soy peptone 0.4%, tryptone 0.4%, yeast extract powder 0.4%, sucrose 1.2% and distilled water 100 mL. The viable count of YE4 with modified YPD can reach 5.01 ×108 CFU/mL, which is more than basis medium one order of magnitude, also, the OD value is 1.06 times than the basis YPD medium.
