摘要
观察补中益气丸对糖尿病大鼠胃组织MUC1、COX-1、COX-2、cNOS、iNOS不同病程表达变化及其相互作用的影响。大鼠随机分为正常组,模型组和中药组,采用STZ腹腔注射结合乙醇灌胃的方法建立糖尿病大鼠胃黏膜损伤模型,检测胃组织NO、PGE2、6-keto-PGF1α、MUC1、COX-1、COX-2、cNOS和iNOS随病程的表达变化及其相互作用。结果显示,糖尿病大鼠出现胃黏膜损伤和胃肠功能紊乱,胃组织MUC1、COX-2、iNOS发生显著变化,MUC1随病程进展逐渐降低,iNOS和COX-2表现为早期活性降低,晚期活性升高,PGE2、6-keto-PGF1α和NO随病程显著升高,相关分析发现,COX-1与cNOS呈显著正相关,与iNOS呈著负相关。补中益气丸能调节MUC1、COX-2、iNOS的表达及其相互作用,阻止胃黏膜损伤。结果表明,STZ腹腔注射结合乙醇灌胃的方法可成功诱导糖尿病大鼠胃黏膜损伤模型,iNOS与COX-2可能为相互协作和相互介导的关系,在糖尿病胃黏膜损伤方面起重要作用,补中益气丸通过升高MUC1、降低iNOS和COX-2的表达及其相互作用,阻止胃黏膜损伤和糖尿病的发展。
To observe the effects of buzhong yiqi pills on the expression of MUC1,COX,NOS and their interaction in gastric tissue of diabetic rats. The rats were divided into 3 groups randomly, the normal group,model group and Chinese medicine group. The diabetic rat gastric mueosa dam- age model was induced by intraperitoneal injection of streptozotocin (STZ) combined with orally administered ethanol. Stomach, pancreas, spleen were sampled for histological examination, ex pression of NO, PGE2,6-keto-PGF1 α, MUC1,COX-1 ,COX-2, eNOS and iNOS gastric tissue in were detected. The resuhs showed,diabetes rats displayed gastric mucosa injury and disorders of gastrointestinal function,gastric tissue MUC1 ,COX-E,iNOS changed significantly, MUC1 gradu- ally decreased as the disease progression,iNOS and COX-2 expression decreased early, and then increased. PGE2,6-keto-PGF1α and NO gradually increased as the disease progresses, correlative analysis showed that COX 1 was positively correlated with eNOS,and iNOS showed significant negative correlation. Buzhongyiqi pills could regulate the interaction and expression of MUC1, COX-2 and iNOS to prevent gastric mucosal injury. It is concluded that STZ intraperitoneal injection combined with ethanol method can induce diabetic rat gastric mucosa damage model, iNOS plays an important role in diabetic gastric mucosal injury,iNOS and COX-2 may be a relationship of mutual cooperation and mutual mediated,Buzhongyiqi pills may through elevated the expres- sion of MUC1 ,decreased the expression of MUC1 ,COX,NOS and their interaction in gastric tis- sue,prevent gastric mucosal injury in diabetes mellitus and diabetic development.
