一种由叶黄素浸膏纯化叶黄素酯的方法研究 被引量:3

摘要 叶黄素是一种广泛存在于花卉、蔬菜、水果中的天然植物色素,其具有抗氧化、减少癌症的发生和发展、增强人体免疫力和预防老年黄斑变性等作用。叶黄素作为一种天然的抗氧化剂和着色剂,在食品、化妆品、医药及饲料等方面得到广泛应用。叶黄素在植物体中主要是以酯的形式存在,研究证实叶黄素酯可经有效水解被人体直接吸收,并且在相同的条件下叶黄素酯的稳定性要优于叶黄素,因此研究制备高纯度叶黄素酯的方法具有广阔的前景。本文以叶黄素浸膏为原料,通过单因素及正交试验优化了叶黄素酯的纯化方法。结果表明,最佳工艺条件为:采用乙酸乙酯:无水乙醇为1∶1的混合液、搅拌时间为30min、洗涤次数为4次、料液比为1∶1.1(g∶g)。在此条件下,叶黄素酯的得率为48.1%,其叶黄素酯含量可以达到870g/kg。 Lutein is a natural pigment widely existed in flowers,vegetables and fruits.It has many functions,such as oxidation resisting,cancer reducing,human immunity enhancing,and the occurrence and developmental senile macular degeneration preventing.Lutein,as a kind of natural antioxidants and coloring agent,has widely used in the food,cosmetic,pharmaceutical and feed industry.Lutein mainly existed in the form of ester in plant.Researches show that lutein ester can efficiently be hydrolyzed by human body and be directly absorbed.Lutein ester has the higher stability than lutein under the same conditions,so the method of producing high-purity lutein ester has broad prospects.Lutein extract was used as raw materials to study the purification of lutein esters through single-factor experiments and optimized by orthogonal test.The results showed that the optimal conditions were as follows:using mixed solution of ethyl acetate and anhydrous ethanol (1 ∶ 1),mixing time 30min,washing for 3 times,solid to liquid ratio of 1 ∶ 1.1 (g:g).Under these conditions,the yield of lutein esters was 48.1%,the content of lutein esters was 870g/kg.
