粒毛盘菌突变株油脂GC-MS分析及其亚油酸甲酯的纯化

摘要 亚油酸甲酯具有较强的触杀、产卵抑制和驱避作用。对一株粒毛盘菌产油脂突变株Z60胞外油脂进行了GC-MS分析,其中亚油酸甲酯(C18:2)占总脂肪酸甲酯质量分数的32.7%。以亚油酸甲酯含量为指标,在单因素基础上,采用正交实验对亚油酸甲酯的纯化条件进行了优化,最佳条件为3.0 g粒毛盘菌胞外油脂,6.6 g尿素,16.5 mL甲醇溶液,-5℃包合18 h,亚油酸甲酯纯度为50.1%。进一步利用硝酸银薄层层析法对其进行分离,分离后亚油酸甲酯气相色谱纯度提高到96.7%。 Methyl linoleate has strong bioactivity of touch kill, oviposit inhibition and repellency. Gas chromatography-mass spectrometry analysis of extracellular lipids produced by Lachnum mutant strain Z60 was performed and the results indicated that the content methyl linoleate was 32.7%. Based on the single factor experiment, the conditions of purification of methyl linoleate were optimized by orthogonal test. The results were as follows: fatty oils 3.0 g, urea 6.6 g, methnol 16.5 mL, -5℃ inclus 18 h. Under this condition, the purity of methyl linoleate reached 50.1%. Furthermore, separation of methyl linoleate linoleate reached 50.1%. Furthermore, separation of methyl linoleate was up to 96.7%.
