摘要
目的:建立胰蛋白酶酶解法制备蛤蟆油蛋白的最佳工艺。方法:采用单因素和正交试验对蛤蟆油蛋白提取的条件进行优化,高效液相凝胶色谱法考察蛤蟆油蛋白的酶解条件,优选出制备蛤蟆油蛋白的最佳工艺。结果:最佳工艺条件为柠檬酸pH4.5,料液比为1:100(g/mL),浸提时间为12 h,胰蛋白酶与底物比为1:100(g/g),酶解时间为12 h。结论:验证试验结果表明,蛤蟆油蛋白提取率为80.66%,蛋白含量为45.95%。
Objective: To establish the optimal preparation technology Oviductus Ranae protein by using trypsin. Methods: Monofactor test and the orthogonal experiment were developed to determine the optimum extraction process, investigate the trypsin hydrolyze conditions by high performance liquid gel permeation chromatography. Results: The optimum preparation technology was obtained as follows: citric acid pH was 4.5, material/liquid ratio was 1 g:100 mL, extracted 12 h, trypsin/substrate ratio was 1 g:100 g, hydrolyzed 12 h. Conclusion: Verifying test results show that the Oviductus Ranae protein yield was 80.66%, protein content was 45.95%.
